How to Pipette PCR Master Mix for Increased Accuracy in qPCR Results

Pipetting technique can influence qPCR assay results. PCR Master mix is routinely used during qPCR set-up but can be challenging to pipette accurately. In this study, we determined the best pipetting technique for pipetting Master mix in qPCR assays. We tested forward and reverse pipetting techniques, the type of pipette tips, pre-wetting of the pipette tip, and the use of electronic pipettes and identified best practices for obtaining accurate qPCR results.

Improving Accuracy Serial Dilutions Using the Electronic Pipette

Serial dilutions are an essential component of dose response curves in drug discovery, limit testing, standard curves for qPCR based assays, and calibration curves in diverse assays. Accuracy in serial dilutions is an important factor that can significantly impact results. In this study, we investigated the use of electronic pipettes for the preparation of serial dilutions. The results demonstrate that use of electronic pipette for serial dilution gives better accuracy in sample concentrations and has reduced error.

Application Note: Pipette Calibration and Standards

Pipetting results are critically depending on the maintenance and calibration of the pipette. The most import International Stardardization Organization (ISO) standards related to pipetting are ISO8655 and ISO17025 and these standards set the guidelines how and in which conditions calibration should be performed.
