Overview
The iQue® analysis is extremely fast - it takes just five minutes to measure a complete 96-well plate. The bottleneck tends to be the sample preparation. Assay setup for flow cytometry consists of many pipetting steps - cells are stained with different dyes and, in the case of iQue® kits, beads that bind secreted proteins are also added to the samples. Thus, despite the iQue® TCA kit being a one-wash assay, the sample preparation is rather laborious and can take hours. The protocol includes three 1-hour incubations and multiple pipetting steps.
