Stem cells can differentiate to several cell types:

  1. Pluripotency stem cells are from embryonic cells or targeted cells that can be reprogrammed to iPSC. Both cell types have self-renewal properties, and can stay undifferentiated for at least 50 passages.
    They can differentiate to each cell from the three lineages of the embryonic germ layer — endoderm, mesoderm, and ectoderm, and  can be used to regenerate tissue in practice.
    For these cells, scientists need a serum-free medium with controlled levels of growth factors to keep cells undifferentiated for as long as possible. As the serum is eliminated from the medium, scientists must add an attachment matrix or carrier (animal source or defined) to the culture to allow the cells to expand.
  2. MSCs are multipotency cells from connective tissues of the body. They have the ability to stay undifferentiated for up to six passages, and can spontaneous differentiate to one of three cell types — chondrocytes, osteocytes, and adipocytes. The cells can be differentiated to other cell types by using specific growth factors. The media we offer keeps cells undifferentiated for at least six passages (depending on the cell source) and allows fast differentiation when needed. As serum is eliminated from the medium, you can add attachment factors or microcarriers.

Sartorius offers media for pluripotent stem cells (PSCs) and mesenchymal stromal cells (MSCs), as well as ancillary products for the culture process from isolation to expansion and differentiation finished with cryopreserved cells.

Media can keep PSCs undifferentiated for at least 50 passages and MSCs for at least 6 passages.

  • PSCs: One ready-to-use bottle of NutriStem® hPSC XF Media.
  • MSCs: Basal media with supplements. As the media is serum-free, it needs to be supplemented with attachment matrix. You can supplement PSCs with laminine and vitronectine, and MSCs with fibrinogen and fibronectin.
  • Detachment: A peptide of recombinant trypsin with or without EDTA.
  • Differentiation: Kits for MSC to chondrocytes, osteocytes and adipocytes.
  • Cryopreservation: Medium based with 10% DMSO and salt based with 5% DMSO.

Totipotent stem cells: Totipotent (omnipotent) stem cells can differentiate into any of the 220 cell types found in an embryo as well as extra-embryonic cells (placenta).
Pluripotent stem cells: Pluripotent stem cells can differentiate into all cell types of the body (but not the placenta). Embryonic stem cells and induced pluripotent cells are under this potency.
Multipotent stem cells: Multipotent stem cells can develop into a limited number of cell types in a particular lineage. MSCs are under this potency.
