Human mesenchymal stem cells (hMSCs) are adult, multipotent stromal cells with the capacity to differentiate into mesodermal lineages including osteocytes, adipocytes, and chondrocytes, as well ectodermal (neurocytes) and endodermal lineages (hepatocytes). HMSCs are self-renewable (up to six passages), easily accessible, and cultures can expand in vitro with exceptional genomic stability. They have an adherent, fibroblast-like (spindle-cell phenotype), shoal-like pattern.
- Positive markers: MSCs have a specific set of cell surface markers (CD73, D90, CD105). HMSCs are non-hematopoietic.
- Negative markers: MSCs lack expression of CD14, CD34, CD45, and human leucocyte antigen-DR (HLA-DR).
Mesenchymal stem cells (MSCs), also referred to as mesenchymal stromal cells, are adult stem cells capable of self-renewal and multilineage differentiation. MSCs can differentiate into cells of mesodermal lineages (adipocytes, chondrocytes, and osteoblasts) and non-mesodermal lineages (ectodermal and endodermal, including neuronal cells, cardiomyocytes, hepatocytes, and epithelial cells). The plasticity of MSCs confers benefits in tissue regeneration. MSCs can inhibit the immune system, promote cell survival, and induce angiogenesis among others pleiotropic activities. The immunosuppressive role of MSCs is particularly interesting for clinical use because it confers resistance to rejection by the host immune system after transplantation.
There are known protocols for isolating MSCs from different tissues. The protocols are well-published and the cells can usually be harvested in aesthetic clinics (e.g., during liposuction) and orthopedic wards. The stages include lysis of cells (mainly fibroblast), connective tissue digestion, pull down of MSCs, and seeding.
NutriStem® MSC XF Basal Medium is suitable to use from the earliest stage. The medium should be used after the lysis step and before seeding to wash the cells. The medium should be equilibrated at the incubator. The complete medium is used in the later stages of isolation, and in the proliferation and seeding of the MSCs. You can obtain MSCs from easily accessible sources using minimally invasive methods, and can rapidly expand them in large-scale clinical use for patient-specific, autologous therapies.
