Viruses are biological entities that replicate only within living cells. Most viruses can be grown in cultured cells such as MDCK, Vero, HEK, or insect cells. Historically embryonated hen's eggs were used to produce viruses before the industry moved to cell culture models.
The cell culture medium used to culture a virus greatly depends on the requirements of the host cell line. Specialty media provide the optimum quantity of nutrients to allow for cell growth and support virus infection and production. Among the human cell lines, HEK293 is widely utilized in viral vector production due to its high transfectivity, rapid growth rate, and ability to grow in a serum-free, suspension culture.
HEK293 cells are easy to grow and maintain in culture. They are also highly amenable to transfection, delivering excellent transfection efficiencies. Thus, HEK293 cells are a preferred cell line for transfection experiments to produce gene therapies or gene-modified cell therapies.
