Pre-Packed Chromatography Columns

Rapid screening of chromatographic conditions is necessary to identify the best purification conditions. Scouting of conditions can be achieved on lab-scale chromatography systems or automated robotic systems. Especially novel functionalities like mixed-mode chromatographic materials require robust screening tools for process development.

Find the Right Solution for Your Chromatography Process

PRC Pre-Packed Columns and Robo Columns for Screening are robust tools for quick purification development. Both products come pre-packed with ion exchange, mixed-mode, and affinity chromatographic materials.

Robo Columns for Screening

Robo Columns for Screening are supplied in strips of 8 pre-packed units. The 200 µL units are suited for screening purification conditions and selecting chromatographic materials. The 600 µL units are recommended when longer residence times above 4 min are needed (e.g., evaluation of dynamic binding capacities).

For High Throughput Chromatography Sorbent Screening and Optimization

ScreenExpert RoboColumns are miniaturized chromatography columns pre-packed with chromatography sorbents. They are designed for fully automated and parallel chromatographic separations with robotic liquid handling workstations, such as the Freedom EVO® from Tecan.

ScreenExpert RoboColumns are packed with anion exchange, mixed-mode,affinity or any other chromatography sorbents upon request.

Two formats are available, 200 μL and 600 μL, with a bed height of 10 and 30 mm respectively. The 200 μL column volume is recommended for sorbent and process conditions screening, while the 600 μL is used for purification process optimization or when a residence time above 4 minutes is required (e.g., capacity study on mixed-mode sorbents).

The columns are supplied as rows of eight pre-packed units, with two removable silicon cover seals for proper storage. A 96-well array plate is available to arrange up to 96 ScreenExpert RoboColumn units.

Benefits

  • Column format, and high quality packing enables the ability to mimic all the steps of a chromatography separation in dynamic mode. Data obtained are comparable to those obtained in larger columns
  • Miniaturized format facilitates screening of chromatography sorbents, reduces sample consumption, and saves time
  • Perfectly fitted to Design of Experiment (DoE) requirements, allowing a large amount of experimental work in a minimum of time
  • 96-well format allows easy integration with automation systems for High Throughput Process Development (HTPD)
  • Multiple parameters can be tested in parallel, e.g., residence time
  • Reusable
  • Flexible use: the user makes column selection when needed

Handling

ScreenExpert RoboColumns are designed for robotic handling.

When using the high-throughput process development approach for ScreenExpert RoboColumns, it is suggested to use Design of Experiments (DoE) criteria for the experimental set-up. This enables to screen many different chromatographic parameters such as resin type, pH, conductivity / ionic strength, etc., efficiently and simultaneously.

For experimental set-up and the configuration of the 96-well array plate, the Te-Chrom Wizard is recommended. The Te-Chrom Wizard is a dialog-based graphic user interface offered by Tecan and based on the Freedom EVOware software. The Te-Chrom Wizard was developed for the use of ScreenExpert RoboColumns and provides the possibility to configure hardware, plate layout, and the chromatographic process as well as to set all process relevant chromatographic parameters, e.g., flow rate, volume without direct script writing.

"Salt-Tolerant" Anion Exchange Chromatography Sorbent

HyperCel STAR AX sorbent is composed of a rigid cellulose matrix that has excellent flow properties and generates low backpressure. It provides:

  • High dynamic binding capacity at short residence time
  • Use in either capture, intermediate or polishing steps
  • High dynamic binding capacity is maintained up to 15 mS/cm conductivity
  • Direct scale up to pilot or production scale columns

Rapid screening and condition optimization in the 1 mL PRC column enables rapid selection of the appropriate separation conditions. Once the separation conditions are selected on a 1 mL PRC column, the conditions of use can be optimized in a 5 mL PRC column.

Two or more columns can be connected in series to increase the column height and more closely model real conditions in pilot scale or for scale down applications. Further scale up can be achieved with minimal re-optimization by packing Pall LRC empty glass columns.

PRC Prepacked Chromatography Columns with MEP, HEA, and PPA...

MEP, HEA and PPA HyperCel™ sorbents exploit unique selectivity of robust synthetic ligands to capture proteins or separate them from contaminants. They provide a unique separation mechanism different from conventional methods. All ligands operate predominantly by hydrophobic interaction. This “HIC-like” interaction typically takes place without the addition of lyotropic salt. This prepacked column format conveniently enables rapid screening and condition optimization.

Rapid selectivity screening of MEP, HEA, and PPA HyperCel mixed-mode sorbents

  • Convenient – Ready-to-use 1 mL and 5 mL prepacked columns.
  • Easy to use – Direct connection to commonly-used laboratory chromatography systems.
  • Efficient – High packing efficiency (> 2,500 plates/meter).
  • Consistent – Screen mixed-mode chromatography sorbents under reliable and reproducible conditions, and guarantee performance run after run.

Mixed-mode Chromatography Sorbents for Antibody Capture and “No Salt” Hydrophobic Interaction

HyperCel sorbents exploit unique selectivities of robust synthetic ligands to capture proteins or separate them from contaminants. They provide a unique separation mechanism different from conventional methods. All ligands operate predominantly by hydrophobic interaction. This “HIC-like” interaction typically takes place without the addition of lyotropic salt.

Rapid screening and condition optimization in the 1 mL PRC column enables rapid selection of the appropriate chemistry. Once the chemistry is selected on a 1 mL PRC column, the conditions of use can be optimized in a 5 mL PRC column in doubling the height.

Two or more columns can be connected in series to increase the column height and more closely model real conditions in pilot scale or for scale down applications.
