When developing an ion exchange (IEX) strategy to purify a new molecule of interest, it is important to optimize the capture and elution conditions to ensure efficient separation. In this application note, we describe the use of a linear elution gradient on Sartobind® Q Lab to establish the relative ionic strengths required to isolate four different proteins. The determined ionic strengths were subsequently applied in a multi-step gradient, to improve separation resolution and purity. With Sartobind® Lab units, optimized IEX strategies can be carried out using Luer lock syringes. This enables faster purifications by eliminating the additional active working time required to run FPLC systems.
