Application Note: Facilitating Rapid and Accurate Cell Line Development

Authors: Daryl Cole, PhD, Darius Wilson, PhD, Nicola Bevan | Last Updated: 17/04/2023

Overview

Generating biologics often requires development of cell lines capable of producing the molecule of interest. Manufacturing a cell line that can be successfully cultured, is robust, and produces the biotherapeutic required at a high enough yield are all key components to a successful cell line development (CLD) workflow.

CLD techniques, such as limiting dilution (LD), traditionally require large quantities of reagents and consumables to achieve single cell seeding as required to create new cell lines. CellCelector Flex mitigates this issue by monitoring clones and ensuring monoclonality from the outset. This application note outlines the utility and value of the CellCelector Flex at key stages of the cell-line development workflow.

Key Takeaways

  • How to reduce the time and resources to successfully produce high quality clones with guaranteed monoclonal lineage.
  • The advantages to the nanowell approach - such as reductions in growth media requirements, space required for plates, time to culture and increases in cell health, expansion potential and titer.
  • The method for characterizing clonal yield potential at very early stages of CLD using a bead-based secretion assay in combination with nanowell technology.
