Semi Automated Workflows for High Quality iPSC Culture | Application Note

Authors: Daryl Cole Ph. D., Senior Scientist, Sartorius  | Last updated: July, 2026

Resource overview

Induced pluripotent stem cells (iPSCs) are a powerful tool for advanced therapies and personalized medicine but are difficult to culture due to their sensitivity and risk of spontaneous differentiation.

To enable evaluation of iPSC pluripotency, this application note describes a semi automated workflow to improve iPSC expansion, selection, and characterization. An automated single-cell isolation platform facilitates gentle, targeted passaging, purification of mixed cultures, and generation of monoclonal spheroids from single iPSCs.

A live-cell imaging system supports continuous, non-perturbing monitoring of growth and morphology. Whilst a high-throughput cytometry platform, provides multiplexed viability and pluripotency marker analysis, confirming maintained stemness.

Key takeaways:

  1. Comparison of Manual and Automated iPSC Passaging
  2. iPSC Culture Purification
  3. Specific Picking and Expansion of Mixed Population iPSCs
  4. Phenotypic Evaluation using Surface Marker Analysis
  5. Monoclonal iPSC Spheroid Generation
