Application Note: Optimizing mRNA Transfection of Human T cells
Overview
Optimizing gene-delivery strategies for T-cells is essential to enhance their therapeutic potential. This study aimed to refine the transfection of human T-cells with mRNA using the jetMESSENGER® transfection reagent.
The specific objectives within this study were to identify culture conditions that maximize cell proliferation, transfection efficiency, and cytotoxic activity against cancer cells. To support T-cell recovery and expansion, the cells were cultured in specialized 4Cell® Nutri-T GMP media, enriched with high-quality cytokines from Sartorius. For kinetic visualization and quantitative assessments, which guided the optimization process, the Incucyte® Live-Cell Analysis System and the iQue® High-Throughput Screening (HTS) cytometer were utilized.
