In vitro reconstructed model of human epidermis containing melanocytes for studying skin pigmentation and lightening

Document type: Poster | Last updated: August, 2025

Overview

Discover a proven in vitro model for fast, reliable screening of skin-lightening and pigmentation-modulating products.

This scientific poster details MATTEK’s 3D human epidermis model incorporating normal human melanocytes from different phototypes (Black, Asian, Caucasian). Cultured up to 21 days at an air–liquid interface, MelanoDerm supports visual, histological, and quantitative assessments of pigmentation changes. Methods include SOLVABLE melanin assays (OD 490 nm) and L* brightness measurements. Example results with kojic acid, magnesium ascorbyl phosphate, and OTC formulations are included.

Applications:

  • Efficacy screening of skin-lightening compounds
  • Comparative phototype response studies
  • Pre-clinical claims substantiation

Key Takeaways

  • Background and rationale for using reconstructed epidermis with melanocytes (MelanoDerm™)
  • Detailed methods: cell sources (NHEK, NHM from different phototypes), air–liquid interface production up to 21 days, and the SOLVABLE melanin assay protocol
  • Results: macroscopic time course of pigmentation, melanin quantitation across timepoints (Day 6/10/14), and effects of test agents (e.g., kojic acid, magnesium ascorbyl phosphate, and several OTC dark-spot correctors)
  • Representative micrographs and figures showing dendritic melanocytes, melanin transfer to keratinocytes, and treatment effects
  • Conclusions and recommended uses for efficacy screening and claims support
