Improving the Efficiency of LV Concentration With Spin Filters Application Note
Authors: Jennifer J Labisch, Andreas Pickl and John S Cashman | Last updated: September 2024
Overview
In this study, we evaluated the performance of Vivaspin® Turbo 15, Vivaspin® 20, and two competitor spin filters for lentiviral vector (LV) concentration. Performance was assessed in terms of process time, impurity removal, and infectious particle recovery. We found the optimal process conditions to be a relative centrifugal force (RCF) of 500 g and a concentration factor up to 10-fold. Vivaspin® Turbo 15 with 100 kDa PES membranes showed the best performance and is recommended for efficient ultrafiltration of LVs and similar-sized viruses.

Key Takeaways
- See how the performance of Vivaspin® Turbo compares to other spin filters when concentrating LV samples
- Understand how process conditions for LV sample concentration can be optimized, taking into account parameters such as RCF
- Recognize that the time to concentrate a sample – a critical consideration in time-sensitive workflows – varies between different spin filters
- Identify which spin filters offer the highest rates of infectious LV particle recovery and impurity removal
