Monolithic Columns as Downstream Processing Solutions for Lentiviral Purification
Lentiviral vectors are efficient tools for transfer and stable integration of large gene inserts into the cell genome, however one of the key challenges remains downstream purification of lentiviruses. This poster outlines the use of CIM® monolithic columns as downstream processing solutions for lentiviral vectors. Various chemistries were tested, with two showing promising results on CIMmic® columns and successfully upscaled to CIMmultus® monoliths.
Key Results:
- Three CIM® monolithic chemistries were identified as promising for lentiviral purification
- Two chemistries showed improved recovery rates when tested on CIMmic® columns and upscaled to CIMmultus® monoliths
- MALS was used for screening of lentiviral vector elution during preparative runs. ddPCR, and infectivity recovery tests were used for analytics
