Comparability Study Between Ion-Exchange Monolith and Affinity Resin For Purification of AAV8

The goal of every AAV downstream process is to achieve high recovery while delivering a pure, potent product, so choosing the right media is vital. The AAV capture step is either affinity or cation exchange chromatography, which both concentrate the vector and remove impurities. The capture step eluate is then further processed to enrich full AAV capsids. For the polishing step, CIMmultus® QA HR, a monolith-based anion exchanger, is widely used. Since the polishing step relies on only small charge differences between AAV capsids, any process-induced heterogeneity or charge modulation of the capture eluate will diminish the separation efficiency and affect the step’s robustness.

This poster presents a side-by-side comparison between the CIMmultus® SO3 monolithic column and a commercially available affinity resin that binds multiple AAV serotypes. Both chromatography columns were evaluated for process and step recoveries, impurity reduction, product capacity, and processing time. The results shown are based on two parallel experiments for each capture step approach.
