Continuous Capture of Adeno-Associated Viruses Using Resolute^® BioSMB

In this proof-of-concept study, we used continuous multi-column chromatography to improve the performance of an AAV capture operation on Affinity Resin. Optimal conditions for continuous bind-and-elute processing were established using traditional breakthrough curve experimentation. These conditions were then tested on the Resolute^® BioSMB PD system, and increased performance was observed. Additionally, the BioSMB operations demonstrated reproducible elution profiles, product purity, and high yields, comparable to those of the AAV batch process (118%).
