The Octet® R2 System makes quality protein analysis affordable and flexible. Priced for startup academic and biotech labs, low volume users are no longer bound by the trade-off between cost and performance when choosing a label-free assay system. The 2-channel Octet® R2 system offers an advanced, fluidics-free approach to protein analysis, with a wide variety of off-the-shelf Dip and Read biosensors for binding kinetics and quantitation analysis. Octet® R2 provides a low-cost entry point for labs with lower throughput needs, while providing the flexibility to field-upgrade to 4 or 8 channels anytime the need arises.

The Octet® R2 System characterizes protein-protein and protein-small molecule binding interactions for early research and discovery. The multipurpose system also provides quantitative information by measuring active protein concentrations — even in complex mixtures like cell culture supernatants and lysates. With sensitivity for small molecule analysis, the Octet® R2 instrument enables the measurement of interactions between small molecules down to 150 Da in size, and their larger binding partners to determine accurate kinetics rates.

The Benefits of the Octet® R2 Protein Analysis System

Kinetic Analysis

The Octet® R2 System monitors binding events in real time to calculate on-rates (ka) of the association, off-rates (kd) of the dissociation, and affinity constants (KD). The system's two channels can be used independently to measure samples for screening purposes, or in tandem, pairing the sample read with a dedicated reference for high-quality kinetic characterization. 

Samples are loaded in standard microplates, eliminating fluidic line maintenance and expanding the number of samples that can be run in an assay. Pre-defined templates in Octet® BLI Discovery software streamline setup prior to running an assay and minimizes training needs. The software also offers users a range of parameters and metrics for analyzing acquired data in a powerful yet intuitive software interface.

Quantitative Measurements

The 2-channel Octet® R2 system directly measures the presence of specific proteins and other molecules in solution with minimal interference from complex matrices. Accurate and reproducible concentrations can be determined in as little as 2 minutes per sample using a simple, one-step assay.

Sample Plate Cooling for Temperature-Sensitive Proteins

The sample plate temperature can be controlled from 15–40°C, which enables reliable kinetic determinations at different temperatures for temperature-sensitive proteins. Additional advantages of sample cooling include the ability to rapidly determine binding rate constants at multiple temperatures to extrapolate thermodynamic measurements. The Octet® R2 system’s two channels can be used independently to measure samples for screening purposes or in tandem, pairing the sample read with a dedicated reference for high-quality kinetic characterization.

Biosensor Re-racking

The Octet® R2 system provides the ability to re-rack biosensors at the end of an experiment. Biosensor re-racking provides enhanced flexibility in loading ligands on biosensors and reduces operational costs when biosensors are loaded with precious target reagents
