Expression Vector
Expression vectors are important tools in the production of proteins. A well-designed expression vector results in enhanced, stable expression and increased secretion.
Cellca Expression vector systems are designed to optimize expression of the protein of interest and minimize any unfavorable impacts of cell growth on the CHO DG44 cells used. These vectors, which are based on the dihydrofolate reductase (DHFR) amplification system, have been constructed using elements that offer the greatest flexibility in operations.
DHFR is expressed only in very low levels, which results in high selection stringency and increases the probability of isolating high-producing clones after the selection process. These DHFR selection systems are designed to work with monomeric and dimeric products.
Low levels of DHFR used for clone selection eliminate the need for methotrexate (MTX)-based amplification. Additionally, no methionine sulfoximine (MSX) and antibiotics are required.
Optimized 5‘ and 3‘ Untranslated Regions (UTRs) regulate mRNA stability and subcellular localization. This results in improved, stable expression as well as optimal signal peptides for enhanced secretion.
Cellca Expression Vector
With the Cellca Expression Vector, genetic elements of the expression cassette have been carefully selected and the distance between genes has been optimized. DHFR selection systems are designed to cover monomeric and dimeric products.
The Cellca Expression Vector utilizes S/MAR elements and proper UTR sequences for enhanced, stable expression as well as optimal signal peptides for increased secretion.
