New capability in the Generation 2 system, enables clone, media and feed screening and investigation of process parameters under perfusion mimic conditions in Ambr^®️ 15 microbioreactors. Here, we detail two different experimental methods, using cell settling and centrifugation steps for cell retention. Set-up, daily cycle of steps, duration of settling events and centrifugation speed/ times are described. Growth and titre results are compared for an IgG-producing CHO DG44 cell line.
